Use classical NMR relaxation experiments to give insight into the dynamical character of integral membrane proteins
Fall 2022 Semester:
I expressed and purified BCL-XL, a protein important in apoptotic signaling
I also inserted BCL-XL into nanodisks for analyzation with NMR relaxation experiments
I expressed and purified the proteins VDAC-1 and MSP-1D1
Spring 2023 Semester:
I purified VDAC-1 (important in mitochondrial mediated apoptotic signaling) and MSP-1D1 (crucial in the formation of nanodisks, which are used by many in the lab to study proteins in a native-state environment for NMR relaxation experiments)
I also expanded upon laboratory skills that I learned in my first semester
Fall 2023 Semester:
I overexpressed and purified GlpG. Since the purification was not quite satisfactory, I spent the semester developing a purification protocol that would maximize the purity of GlpG. The goal is to reach a purity suitable for NMR relaxation experiments.
Spring 2024 Semester:
I continued to work on developing a purification protocol for GlpG. Anion exchange chromatography, size exclusion chromatography, and cobalt affinity chromatography were all utilized to achieve this.
